It sort of depends upon what you're betting, but the short answer is the Cantor shops. They open lines for every NFL week (for small limits) long before the season starts. FWIW, the Cantor franchise at the Palazzo is the nicest and busiest book in town (at least on weekends). You'll need to pay a cover to get in there on an NFL Sunday.

They used to a few years ago for NFL - when they were still an independent book. Not sure if they still do. I never go there on Sundays.

It's not often I agree w/ JJ's perspective, but I too prefer off-strip - Red Rock to watch a game. Better cocktail service, a little less crowded.
For convenience I bet through Cantor, set my acct up at Palms. I bet using the phone app then just cash out before I leave town. I also like that cantor offers in-game live betting every day.
For hassle-free large bets, LVH hands down...

LVH is a good book. They have some in game wagering. Lots of screens, not too hard to get a seat. Decent cocktail service. Old clientele, cocktail waitresses were prob serving the Rat Pack (not exaggerating). They put the NFL games on in the theatre. It's a cool atmosphere, raffle off free stuff, $2 beers, $1 hot dogs.

rudy just go to the M man.. that place is what you are looking for.. trust me..
if you want to get in and out quick though.. like hop out a cab .. place a wager.. get an awesome line with big money and be back in that cab in 5 mins... Tropicana is Cantor and you are in and out like the fing A-team... also LVH has its own entrance where you walk through one outside door and you are in the book .. it even has parking...

Lagasee does not charge cover to enter.
If you want to sit in one of the seats, I believe it is a $50/minimum per person
