Winning poster: I am planning to donate part of the winnings to Japan Earth Quake Relief. They need all of our help now. Besides that, I want to pay off my education loans. The rest, I am planning to invest in some stocks / ventures.
Japan Earthquake Relief = $100
Halfhearted attempt to payoff education loans = $1000
Hookers/Blow = $38k
Future 10 teamers = $100k

You collect the money in whatever way the book will pay you. Then for that tax year where you collected the money you do an accounting of money lost versus money won in all of your gambling adventures that year and you report any net winnings (the amount above your losses). That's it. The net win is treated as income and you pay taxes on it.
Obviously there is a lot to discuss with an accountant (tax rate, other write-offs, etc.) as those can be substantially helpful but for the purposes of a quick answer, the above paragraph would be it.
